What plans should be prepared before launching a clinical trial?
Preparing a clinical trial involves more than developing the protocol, obtaining the required approvals, or signing agreements with study sites. Before participant recruitment begins, a number of operational documents should also be prepared to define how the project will be conducted and ensure compliance with ICH GCP principles and applicable regulations. Their scope depends on the type and complexity of the study, but their common goal is to ensure high-quality data, participant safety, and efficient project execution.
One of the most important documents is the Project Management Plan, which describes the organization of the study, timeline, roles and responsibilities of team members, progress reporting, and communication between the sponsor, CRO, and study sites. This document provides the foundation for managing the entire project.
A key element of the quality system is the Quality Management Plan, which defines quality assurance principles, monitoring of critical processes, and the implementation of corrective and preventive actions (CAPA). It is complemented by the Risk Management Plan, which identifies risks that may affect participant safety, data quality, or protocol compliance and defines measures to mitigate them.
Another important operational document is the Monitoring Plan, which describes the monitoring strategy, scope of source data verification, visit frequency, and procedures for remote and on-site monitoring. Increasingly, it also incorporates risk-based monitoring (RBM) elements. The Medical Monitoring Plan, in turn, defines medical oversight procedures, adverse event assessment, and cooperation between investigators, the sponsor, and the Medical Monitor.
Effective data management is supported by the Data Management Plan, which describes the processes for eCRF design, data entry, query management, medical coding, and database lock. It is complemented by the Data Validation Plan, which defines data validation rules, quality checks, and procedures for identifying and resolving discrepancies.
Before the analysis of study results, a Statistical Analysis Plan (SAP) is prepared. It provides a detailed description of statistical methods, analysis populations, and the approach to evaluating endpoints. This ensures that analyses are conducted according to predefined rules, increasing the reliability of the results.
An important part of the documentation is also the Protocol Deviation Management Plan, which defines how protocol deviations are identified, classified, and assessed, as well as the actions taken to minimize their impact on participant safety and data quality. The eTMF Plan is also becoming increasingly important. It defines how the electronic Trial Master File is maintained, how documentation is compiled, and who is responsible for keeping it up to date and aligned with the TMF reference model.
Properly prepared plans enable efficient clinical trial management, clearly define the responsibilities of all project participants, and minimize the risk of errors and delays. They form the foundation for successful trial execution and are an important part of preparing the project for implementation, as well as for subsequent audits and inspections.
